ancestors

How do I go about a search for cherokee ancestors in my life line. Some one told me that I could have a DNA test for this.
My wife has pictures of her grandparents, the Richs that seem strange and we don't know how to authenticate.
We live in Asheville and could drive over ant day. I just watched a PBS special and I think my mother was moved to FL.
Any help would be greatly appreciated.
